(PO33) Exploring the Influence of a Novel Endodontic Irrigation Solution on Dentin: Unraveling the Dynamics of Growth Factors and Cytokines

Abstract: Regenerative endodontics involves a complex cascade of cellular events mediated by bioactive molecules involved in directing cellular differentiation. The aim of this study is to evaluate the impact of a new endodontic irrigant composed of 10% citric acid + 1% Chlorhexidine (CACHX) on the release of cytokine growth factors from dentin. With the approval of the ethics committee, 15 single-rooted teeth were standardized to 12 mm and instrumented with reciprocating system files (R50) and subsequently divided into 5 groups (n=3): sterile water (control); CACHX; citric acid 10 %; EDTA 17%; and sodium hypochlorite 2.5%.The samples were immersed in 1 mL of saline solution and incubated at 3º C for 1, 7 and 14 days. Using XMAP technology (Luminex), the concentrations of 27 biological mediators were analyzed. The analytes, that were quantified in a Bioplex MAGPIX system (Biorad, California, USA), showed constant release over time, the vascular endothelial growth factor (VEGF) had low concentrations for all irrigators. CACHX released the highest amount of platelet-derived growth factor (PDGF) in 24 hours. For all solutions tested, TNFα, IL1β and IL6 had similar concentrations, while IL8 had higher concentrations with 17% EDTA on 1 and 7 days. Through the results obtained, dentin shows a dynamic characteristic as a source of bioactive molecules that can be released differently depending on the irrigant. CACHX favors the release of VEGF, PDGF and FGF, in addition to inflammatory modulating cytokines with comparable action to the use of EDTA and citric acid.
