(PO26) Effect of Polydatin on the Viability and Odontogenic Differentiation of Human Dental Pulp Stem Cells

Abstract: Various materials have been used to promote human dental pulp stem cells (hDPSCs) differentiation, aiming to produce dentin bridge formation at the coronal most part of the pulp, with less-than-optimal results. A material capable of promoting odontoblastic differentiation while reducing the inflammatory process might prove advantageous in various dental pulp regeneration/repair procedures. Polydatin (PD), a naturally occurring compound found in traditional Chinese medicine, is known for its anti-inflammatory and osteogenic properties. PD has shown encouraging results in promoting stem cell osteogenic differentiation. The effect of PD on hDPSCs in terms of cellular viability, alkaline phosphatase production, and quantitative reverse transcription polymerase chain reaction (RT-qPCR) for odontogenic and osteogenic markers production, including dentin sialophosphoprotein (DSPP), dentin matrix protein-1 (DMP-1), Runt-related transcription factor-2 (Runx-2), osteocalcin (OC), and alkaline phosphatase (ALP), was evaluated. In addition, Alizarin red staining for mineral deposits was performed. The viability of hDPSCs in the presence of 0.01 µM and 0.1 µM PD was significantly higher on days 3 and 7, respectively. Additionally, the ALP activity of hDPSCs was significantly increased with 0.01, 0.1, and 1 µM of PD. Furthermore, PD promoted an increase in the size of mineral deposits. On the other hand, increased expression of messenger RNA (mRNA) for the following markers: ALP, OC, ON, OP, RUNX-2, DSPP, and DMP-1 was observed in the presence of PD; however, the difference was not statistically significant. PD promoted the expression of markers associated with odontogenic differentiation and mineralized tissue deposition in hDPSCs.
