(PO35) Assessing Biodendrimer Resveratrol Effects on Lipopolysaccharide-Induced Cytokine Upregulation in Human Pulp Cells

Abstract: Vital pulp therapy (VPT) aims to preserve pulp tissues and maintain tooth function, often alleviating dental pain. However, persistent pain may necessitate more invasive treatments like root canal therapy (RCT). The current VPT protocol lacks drugs that could enhance tissue healing and improve post-operative relief. Resveratrol (RESV), a safe agent with anti-inflammatory, antimicrobial, and neuroprotective effects, holds promise for VPT. This study aims to establish the non-toxic Dendrimer-RESV dose and evaluate its anti-inflammatory efficacy in VPT. DPSCs and DPCs from healthy adult teeth underwent up to 10-day treatments with varying RESV (0.001-10µM), LPS (0.001-100µg/ml), or LPS + RESV doses. Cell viability was assessed using the MTT assay on days 3, 7, and 10, and cytokine expression was evaluated through qRT-PCR on day 2. All doses of RESV had no impact on DPSC viability, but the highest dose of Dendrimer-RESV significantly reduced DPSC viability. Quantitative RT-PCR analysis showed that LPS induced upregulation of IL1β, IL6, IL8, CCL2, and TNFα in both DPSCs and DPCs. RESV treatment in DPSCs showed a significant decrease in LPS-induced IL-1β and IL-6 upregulation (p < .01). However, Dendrimer-RESV treatment in DPCs decreased LPS-induced IL8, CCL2, and TNFα upregulation only. Dendrimer-RESV demonstrated the ability to mitigate LPS-induced cytokine upregulation, suggesting its potential as an inflammatory response modulator. Further research is necessary to confirm RESV's viability as a VPT supplementary component.
