(PO118) Effect of the Bacterial Microbiome on Host Response in Symptomatic Irreversible Pulpitis

Abstract: Dental caries and microbial invasion into the pulp space can cause symptomatic irreversible pulpitis (SIP), an inflammatory response of host cells within the pulp. The interaction between the bacteriome and the dental pulp cells in the SIP condition is not well understood. Analysis of the transcriptome of pulp tissue from teeth with SIP can describe the mechanism of pulp cell reaction to the SIP-specific bacteriome. Additionally, microbial species within the pulp tissue can be identified by their specific RNA sequences. These data can be obtained simultaneously from a single sample using dual RNA-Seq technology. The aim of this study was to examine the differences between the pulp tissue of teeth clinically diagnosed with SIP and the pulp from non-carious teeth, regarding the pulpal microbiome and the host cell response. Pulp tissue was collected from 10 extracted non-carious impacted third molars and from 20 teeth clinically diagnosed with SIP. Total RNAs were extracted from the combined sample of each group. Dual RNA-Seq technology was used to determine the transcriptomes of dental pulp cells and the pulpal microbiome. Gene profiles of the transcriptome of host cells and the pulpal microbiome were analyzed. The data revealed drastic changes in the transcriptome as well as in the microbiome in the SIP condition. These observations offer insights into the host-bacteria interaction in SIP and may contribute to future advancements in the treatment of SIP.
