(PO19) Citocompatibility Analysis of six Different Endodontic Sealers

Abstract: Endodontic sealers traditionally seal dentinal tubules, creating a homogenous interface between the obturation material and the dentinal walls. Bioceramic sealers offer potential advantages due to their bioactivity. Successful osseous healing post-endodontic therapy relies heavily on osteoblast differentiation and activity^1,2. The aim of this study is to evaluate the cytocompatibility of six different endodontic sealers through biological characterization. Materials included AH Plus (Dentsply), Sealapex (Kerr), Ceraseal (META Biomed), BioRoot RCS (Septodont), EndoSequence BC Sealer (Brasseler), and Tubli-Seal (Kerr), along with an MTT reagent, a CO_2 incubator, an inverted microscope, a Neubauer Chamber, sterile Eppendorf tubes, and sterile Falcon tubes. The experiment exposed a fibroblast cell line (L929 ATCC-CCL-1) to various sealer concentrations for 24, 48, and 72 hours. Cell viability was determined through an MTT assay measuring metabolic activity³. Statistical analysis involved one-way ANOVA with Bonferroni and Tukey’s post hoc tests. Bioceramic sealers exhibited greater biocompatibility with the cell line, with EndoSequence BC Sealer showing the highest compatibility (98%), followed by Ceraseal (92%) and BioRoot RCS (86%). Tubli-Seal (57%) and AH Plus (68%) had the greatest impact on reducing cell growth, while Sealapex (49%) was considered the most cytotoxic, although it showed good compatibility at 24 hours. All sealers demonstrated high cytocompatibility percentages at 24 hours. Only bioceramic-based sealers maintained high cytocompatibility at 24, 48, and 72 hours of the study. These findings suggest that bioceramic sealers may offer superior cytocompatibility compared to other endodontic sealers.
