(PO37) Influence of Hydraulic Calcium Silicate Sealers on Human Dental Pulp Stem Cells - In Vitro Study

Abstract: To investigate the influence of BioRoot RCS, BioRoot Flow, and AHPlus Bioceramic sealers on the behavior of Human Dental Pulp Stem Cells (HDPSCs) in vitro. Discs measuring 5 x 2 mm of BioRoot RCS sealer (Septodont, France), BioRoot Flow sealer (Septodont, France), and AHPlus Bioceramic Sealer (Dentsply Sirona, USA) were produced. HDPSCs were seeded at a density of 5,000 cells/well in a 24-well plate and incubated for 24 hours. Untreated HDPSCs served as the control. The HDPSCs were exposed to the tested materials through direct and indirect contact approaches. Cell proliferation and metabolic assays, alongside phase-contrast microscopic imaging, were conducted following direct and indirect contact approaches at 24 and 48 hours of exposure. BioRoot RCS enhanced cell proliferation of HDPSCs by 50.8% over the control at 24 hours, with an observed 12.5% decrease at 48 hours. BioRoot Flow exhibited a 36.6% increase at 24 hours, but a 41.3% decrease at 48 hours was noted. AHPlus Bioceramic displayed consistent cell proliferation with a 19.3% increase at 24 hours and a further 4.7% increase at 48 hours in comparison to the control. Metabolic activity in a direct approach for BioRoot RCS and BioRoot Flow was initially higher than the control at 24 hours, with a subsequent decrease for both sealers at 48 hours. In contrast, AHPlus Bioceramic showed a marginal increase at both time intervals. Indirect contact assays showed increased metabolic activity for all sealers at both 24 and 48 hours, with the exception of AHPlus Bioceramic showing a slight decrease at 48 hours. All sealers demonstrate favorable in vitro metabolic activity and cell proliferation on HDPSCs.
