(PO78) Comparison of Tissue Dissolution Kinetics between Two Endodontic Irrigation Solutions

Abstract: Endodontic irrigation is a critical component for the disinfection of the root canal system. Traditionally, sodium hypochlorite (NaOCl) and Ethylene-diamine-tetra-acetic-acid (EDTA) have been the irrigation solutions of choice for tissue dissolution and chelation. More recently, Triton^® All-in-One Irrigation (Brasseler) has been well-received as a sole irrigation solution that claims simultaneous organic and inorganic tissue dissolution. The objective of this study is to compare the tissue dissolution kinetics of Triton with NaOCl (6%). Tissue dissolution kinetics was performed with real-time in situ FTIR (ReactIR – Mettler-Toledo). Bovine tissue was suspended in NaOCl/Triton with a ReactIR probe, and FTIR spectra (ICIR software, ver.9.9.1) were collected at one-minute intervals. For the weight-loss method, tissue was weighed (mg) before and after suspension in NaOCl/Triton and measured for up to 5 minutes (n=5). The data were analyzed using Student’s t-test (p < .05). FTIR spectral analysis for amide, bound, and free protein was compared between the two groups. The data showed a significant increase in amide and free protein in the Triton group up to two minutes, and NaOCl group carbonyl bands were seen at 2-minute intervals. The average dissolution weight loss in the Triton group was significantly (p < .05) higher than in the NaOCl group. Within the study's limitations, rapid tissue dissolution was seen with the use of Triton when compared to NaOCl alone. This suggests that Triton is a multifunctional single irrigation solution with more efficacy in tissue dissolution and antimicrobial activity, thus reducing the quantity of irrigation solution leading to reduced chair time. More studies are needed to evaluate cytotoxicity and patient-oriented outcomes.
